Abstract

This experiment was aimed to find some indicators related to primers and genetic variations. Six bread wheat genotypes were selected as the dataset. The RAPD marker technique was used with oligonucleotide primers as marker of study. The goal was to identify factors used for the relationship between primers and their ability to elicit DNA fragments through different genotypes, as well as the relative genetic band abundance among genotypes. After the designed factors, key findings emerged: in relative primer amplification rate, C1 genotype exhibited a higher relative primer amplification rate (RPAR) of fragments from OP-H01 but did not differ from OP-V19 primer. The C2 genotype displayed higher RPAR of fragments from OP-H01 and OP-K01 primers. In the C3 genotype, OP-P04, OP-V19 and OP-K01 primers had high relative primer amplification rate. The OP-P04 primer exhibited highest RPAR compared to other genotypes. In C5 genotype, OP-M14 and OP-H01 primers gave high values. OP-M14 primer was most effective in C6 genotype. Overall, OP-V19 and OP-P04 primers produced the highest number of bands that they were given most variations. C4 genotype exhibited the highest Relative genetic band abundance compared to the other genotypes. The study showed that the OP-H01 primer did not demonstrate genetic variations between the genotypes compared to the other primers. The C1 and C6 genotypes did not show significant interaction with the primers used, thus exhibiting the lowest and highest content of relative genetic band abundance.
